Comment/Description

In this course, basics of fluorescence microscopy and super-resolution microscopy techniques will be covered, with the focus on single-molecule localization microscopy techniques. Two of the most important techniques, dSTORM and DNA-PAINT, will be discussed in detail. While dSTORM is more established and easy to implement, DNA-PAINT is a rapidly developing and versatile technique, capable of reaching superior resolution of single nanometers. Recent developments in cellular DNA-PAINT enable to exploit the full potential of DNA-PAINT for multiplexed imaging in fixed cells, then contributing to a deeper understanding of cellular organization and interplay. Advantages, limitations and experimental parameters optimization of dSTORM and DNA-PAINT will be discussed. In the practical part of the course, we will have a hands-on the multiplexed DNA-PAINT imaging of fixed cells.

Course duration: two days. The course will include lecture/seminar/detailed overview of the experimental setups/hands-on session/data analysis training. Following topics will be included:

* Introduction to optical microscopy and super-resolution techniques
* dSTORM imaging
* DNA-PAINT imaging (including multiplexing and variants of DNA-PAINT)
* Microfluidics technology employed for multiplexed Exchange DNA-PAINT imaging
* Multiplexed super-resolution imaging of fixed cells with 10 nm resolution
* Fluorescence lifetime single-molecule localization microscopy
* data analysis of the collected data using different software packages
* quantitative resolution estimation of the obtained super-resolution images



Important:

If you do not provide this, you will be excluded from courses for the next 12 months, as well as travel grants and other financial support. No exceptions.
