This course will provide basic knowledge and skills on performing electron microscopy of synapses.

Participants will get insights into:



- sample preparation for electron microscopy (due to time limitations partially theoretically)

- Ultrathin sectioning

- Post-staining

- Image acquisition

- Electron-tomography.



In order to prepare inner hair cell ribbon synapses and biological tissues in general for transmission electron microscopy many steps are necessary. The tissue needs to be fixed/immobilized, stained and dehydrated. Moreover, embedding in specific resins is required, followed by trimming steps and ultrathin sectioning. We use this conventional room temperature embedding method to prepare the organ of Corti and/or the anterior ventral cochlear nucleus (AVCN) for electron microscopy. We are interested in the structure-function correlation of the inner hair cell (IHC) ribbon synapses which play a key-role in the hearing process. After passing the IHC ribbon synapses, the signal is transmitted to the AVCN for further processing. We investigate IHC ribbon synapses as well as synapses of the so-called endbulbs of Held in the AVCN and determine parameters such as the number of vesicles, docked vesicles and the vesicle diameter in wild-type or hearing impaired mice using electron microscopy.
